“New Experimental and Data Analysis Methods for Fluorescence Nanoscopy”

Abstract:

 

Light microscopy, and in particular fluorescence microscopy has the ability to image cellular components - down to single proteins - with high specificity in live or intact cells. The continuing development of contrast agents, e.g., fluorescent tags, and imaging modalities such as super-resolution, light-sheet, and hyperspectral microscopy are pushing the boundaries of achievable spatial-temporal-spectral resolution and multiplexing capabilities. These microscopy techniques produce large imaging data sets that are rich in information and these data sets are often not fully exploited.  I will discuss several experimental efforts to improve throughput, multiplexing and spatiotemporal resolution in fluorescence nanoscopy as well as several Bayesian methods to extend resolution and extract kinetic parameters from super-resolution and single particle tracking data.
