Molecular Evolution Protein Engineering and Production facility

The MEPEP facility will be organized in two main subunits: 1) Molecular Evolution and Protein Engineering and 2) Protein Expression, Purification and Characterization. The Molecular Evolution and Protein Engineering part will focus on applying directed evolution techniques for improving protein function and properties. We will achieve this by iterative rounds of mutations, expression and screening of the protein’s property of interest. As one of the main bottlenecks of the directed evolution process is the screening, we will also focus on developing suitable high-throughput detection methods based on flow cytometry and/or microfluidics devices for each property and protein of interest. The Protein Expression, Purification and Characterization subunit of the MEPEP facility will focus on small to mid-scale expression of recombinant proteins in different prokaryotic (E. coli), eukaryotic (S. cerevisiae, P. Pastoris, CHO, HEK) cell lines or in vitro transcription-translation systems. We will develop the protocols based on the commonly used laboratory strains but also work on strain engineering for better expression yields or properties (e.g. different glycosylation patterns).
